The biochemistry and molecular biology of the dihydropyridine-sensitive calcium channel.

چکیده
Calcium channels are known to exist in muscle, neuronal and secretory cells. The 1,4-dihydropyridines are potent blockers of L-type Ca channels, and have been used as specific probes in the study of dihydropyridine-sensitive Ca channels. The receptor for the 1,4-dihydropyridines has been purified from skeletal muscle in order to characterize the biochemistry and molecular biology of the dihydropyridine-sensitive Ca channel. This review summarizes recent findings on the subunit composition of the dihydropyridinesensitive Ca channel, and discusses the structure and possible function of the individual subunits.

The role of calcium channel blockers in wound healing
Wound healing is a natural response to restore the injured tissue to normal. Wound healing is also complicated process involving different cellular, molecular and biochemical mechanisms and various types of cytokines and growth factors. Calcium channel blockers belong to cardiovascular medicine and administrated to treatment of hypertension, angina and cardiac arrhythmia because of vasodilatory...
متن کاملThe dihydropyridine-sensitive calcium channel of the skeletal muscle: biochemistry and structure.
The dihydropyridine-sensitive calcium channel of the rabbit skeletal muscle is the first voltage-gated calcium channel which has been purified and biochemically characterized. The alpha 1-subunit, a 165 kDa protein, of the purified dihydropyridine receptor contains all regulatory sites of a L-type calcium channel and the calcium conducting unit. The purpose of this review is to summarize and di...
متن کاملSynthesis and Effects of Novel Dihydropyridines as Dual Calcium Channel Blocker and Angiotensin Antagonist on Isolated Rat Aorta
Objective(s) Four novel losartan analogues 5a-d were synthesized by connecting a dihydropyridine nucleus to imidazole ring. The effects of 5a and 5b on angiotensin receptors (AT') and L-type calcium channels were investigated on isolated rat aorta. Materials and Methods Aortic rings were pre-contracted with 1 pM Angiotensin II or 80 mM KCl and relaxant effects of losartan, nifedipine, 5a and...
متن کاملMuscular dysgenesis in mice: a model system for studying excitation-contraction coupling.
Muscular dysgenesis (mdg) is a lethal autosomal, recessive mutation of mice. Skeletal muscle from dysgenic mice is paralyzed due to the failure of excitation-contraction (E-C) coupling. Considerable evidence indicates that this failure results from the absence of a specific gene product, the alpha 1 subunit of the skeletal muscle receptor for dihydropyridine calcium channel modifiers. This dihy...
متن کاملSynthesis and Biological Evaluation of New Tricyclic Dihydropyridine Based Derivatives on Potassium Channels
The present study reports a microwave-assisted method for the synthesis of twelve novel tricyclic 1,4-dihydropyridine derivatives in which dimethyl-substituted cyclohexane and / or tetrahydrothiophene rings are fused to the DHP ring. The structures of the compounds were confirmed by spectral methods and elemental analysis. The potassium channel opening effects of the compounds were determined o...
